Akentrogonid rhizocephalans morphologically resembling the genus Thompsonia are revised as a result of examination of new material. The species concerned are all obligatorily colonial and have ovoid or cylindrically shaped externae with a terminal stalk and a much reduced anatomy. A numerical cladistic analysis of all Rhizocephala Akentrogonida using the Hennig 86 program leads to a redefinition of the Thompsoniidae HOeg and Rybakov, 1992. Autapomorphies for the Thompsoniidae are primarily the morphology of the attachment to the host and the total absence of a mesentery. The cladistic analysis refutes that the Thompsoniidae should have a plesiomorphic morphology and branch off very low on the rhizocephalan phylogeny. The family now comprises four genera: Pottsia gen. n. (monotypic), Diplothylacus gen. n. with two species, Thompsonia Kossmann with five species. A revived and redefined Thylacoplethus Coutière includes eight species. The genera are distinguished by the location of the spermatogenic tissue, the site where the eggs are fertilized, the presence or absence of a mantle pore and the way it is formed, the number or absence of oviducts, and the number of cuticular annuli on the stalk. All 16 species, of which six are new to science, are described when necessary, and, if possible, illustrated. A phylogeny for the redefined family is proposed. Thylacoplethus is morphologically closest to the hypothetically ancestral thompsoniid and is likely paraphyletic. The new genus Polysaccus with two species, one of them new to science, and the monotypic genus Pirusaccus Lützen resemble thompsoniids in externa morphology and in being obligatorily colonial.  相似文献

Previous studies have demonstrated that direct or indirect elevation of cAMP levels in cultured amphibian ovarian follicles simultaneously stimulated production of oocyte maturation-inducing steroid (progesterone) by the follicles and inhibited oocyte maturation induced by endogenous or exogenous hormone. The duration of cAMP stimulation influenced arrest and reinitiation of oocyte meiotic maturation in ovarian follicles of Rana dybowskii. Addition of forskolin (adenylate cyclase stimulator) to cultured follicles inhibited both progesterone- and frog pituitary homogenate (FPH)-induced oocyte maturation. Similar inhibitory results were obtained when hormone-treated follicles were cultured in the continual presence of cAMP. Oocyte maturation increasingly occurred in follicular oocytes when cAMP or forskolin addition was delayed following treatment with FPH or progesterone. Transient exposure (6-8 hr) of ovarian follicles to forskolin or cAMP markedly stimulated oocyte maturation as well as accumulation of progesterone as measured by radioimmunoassay within the ovarian follicles. Forskolin was more effective than cAMP, at the dose tested, in stimulating progesterone production and accumulation by the follicles. The data demonstrate that transient manipulation (elevation) of cAMP levels in cultured follicles, without added FPH or steroid, was sufficient to initiate oocyte maturation. Results suggest that, with transient exposure to forskolin or exogenous cAMP, there is a sequential increase and decrease in endogenous cAMP levels in the somatic cells and germ cell components of the ovarian follicle. These changes appear to mediate production of maturation-inducing steroid and secondarily allow its effects on the oocyte to be expressed.  相似文献

The Rhizocephala are considered to be monophyletic due to several synapomorphies in the ontogeny of the cndoparasitic phase. The various types of metamorphosis described in the Rhizocephala are discussed and compared to metamorphosis in the Cirripedia Thoracica and Acrothoracica. In males and females of the suborder Kentrogonida. the cyprid settles and metamorphoses into a new instar, in males the trichogen and in females the infective kentrogon. The kentrogon goes through yet another. incomplete moult associated with the development of the stylet. Within the three kentrogonidan families. the Iernaeodiscid-peltogastrid type of kentrogon differs from the sacculinid type in the mode of attachment to the host. in the complexity of internal anatomy. in the position and penetration of the stylet, and in whether or not the cyprid carapace must be shed prior to penetration of the stylet. In the Akentrogonida metamorphosis never results in a new instar. Where observed (Clistosaccidae and Thompsoniidae). both male and female cyprids settle and penetrate into their substrate (female parasite or new host) with one of the antennules. Using the antennule as a syringe. male cyprids inject spermatogonia while female cyprids injects embryonic cells developing into an endoparasite. By comparison with metamorphosis in the Cirripedia Thoracica and Acrothoracica it is concluded that the presence of a metamorphic moult leading to a post-cyprid instar is plesiomorphic and that the trichogon and kentrogon are homologous with the first metamorphosed juvenile in these outgroups. The abbreviated ontogeny in the Akentrogonida without metamorphic moult and post-cyprid larval instars is considered apomorphic. This contradicts the long-held supposition that the Akentrogonida are the most‘primitive’Rhizocephala and dovetails with new information that this suborder contains many advanced traits. Within the Kentrogonida. the lernacodiseid-peltogastrid type of kentrogon is considered more plesiomorphic than the sacculinid type, which resembles the clistosaccidthompsoniid type in having the antennules involved in the penetration process. The homologization of the kentrogon with a juvenile barnacle indicates that presence of a kentrogon is plesiomorphic within the Rhizocephala and that the Kentrogonida is paraphyletic.  相似文献

Family 3A mammalian liver cytochromes P450 (3A1, rat; 3A3/4, human) catalyze the 6 beta-hydroxylation of endogenous steroids and are steroid inducible. Our recent finding that A6 cells (a toad kidney epithelial cell line) contain corticosterone 6 beta-hydroxylase activity as a steroid-inducible microsomal cytochrome P450 raised the possibility that corticosterone 6 beta-hydroxylase activity in the A6 cells is catalyzed by a member of the 3A family. We found that incubation of A6 cell microsomes from dexamethasone-induced cells with antibodies against family 3A proteins specifically inhibited corticosterone 6 beta-hydroxylase activity. Microsomes from A6 cells analyzed on immunoblots developed with family 3A specific antibodies revealed immunoreactive proteins and treatment of A6 with corticosterone or dexamethasone increased the amounts of 3A immunoreactive protein(s). Furthermore, A6 RNA hybridized with 3A cDNAs on Northern blots and genomic DNA from A6 cells hybridized with a 3A cDNA on a Southern blot. Thus, toad kidney A6 cells express a family 3A P450 that is immunochemically, functionally, and genetically related to the mammalian liver 3A proteins. Prompted by these findings in amphibian kidney, we examined mammalian kidney for evidence of family 3A proteins. Immunocytochemical studies of frozen cryostat sections of normal adult rat kidney incubated with 3A1 antibody showed immunoreactivity only with collecting duct. Immunoblot analysis of human kidney microsomes found three protein bands representing 3A3/4, 3A5, and a 53-kDa Mr protein immunoreactive with human 3A antibody. An unexpected finding was the polymorphic expression of 3A3/4 in human kidney with only one of seven (14%) adult human kidneys tested expressing this protein while 3A5, a protein which is polymorphically expressed in adult human livers, was routinely present in the adult human kidney samples tested. Since human fetal liver contains a family 3A P450 we examined human fetal kidney microsomes by immunoblot analysis with human liver 3A antibody and found expression of a protein tentatively identified as 3A7. Thus, like A6 amphibian cells, family 3A P450 proteins and mRNAs are prominent, functional components in the kidney of mammals, including man.  相似文献

The follicle wall was previously shown to be involved in insulin induction of oocyte maturation in Rana pipiens ovarian follicles. Steroidogenic involvement in insulin induction of maturation was investigated following development of a radioimmunoassay (RIA) for progesterone to measure endogenous progesterone associated with in vitro incubates. Insulin and frog pituitary homogenate (FPH) were both found to elevate progesterone levels significantly in these incubates. FPH was more effective in elevating progesterone levels than insulin and caused progesterone increase of about 2 orders of magnitude greater than insulin. Removal of the follicle wall eliminated the steroidogenic effects of insulin. Considerable interanimal variation was observed in the ability of insulin to induce oocyte germinal vesicle breakdown (GVBD) in intact follicles. The hypothesis was proposed that differences in endogenous progesterone might explain this variation. To test this hypothesis, an experiment was carried out in which hormone production and follicular sensitivity to insulin were simultaneously determined in follicles obtained from the same animals. Results of the experiment show that the ability of insulin to induce GVBD, as indicated by the effective concentration needed for 50% response (ED50), was strongly correlated with the levels of endogenous progesterone as measured by RIA. The results provide direct evidence that insulin's action on the follicle wall involves steroid production. It was thus concluded that increased endogenous progesterone facilitates GVBD induction by insulin. It is unclear how the two hormones interact to produce an enhanced effect, but interactions at the receptor or postreceptor level may be involved. This follicle system may provide important insights into the mode of action and interaction of these two important hormones.  相似文献

Starfish oocytes released from the ovary into sea water during the breeding season underwent spontaneous meiotic maturation. In the absence of calcium ions, follicle cells surrounding the oocyte were dispersed and oocyte maturation did not occur. Immature oocytes in the absence of follicle cells did not mature even when calcium was available. Calcium ions stimulated the release of a meiosis-inducing substance from follicle cells which had been isolated in calcium-free sea water. Increasing numbers of follicle cells produced a parallel increase in the amount of meiosis inducing substance released. Spontaneous oocyte maturation appeared to be initiated and mediated in part by calcium sensitive processes in the follicle cells rather than the oocyte.  相似文献
